چکیده

Radiosensitivity/Ionizing radiation/Cell killing/Human T-lymphocytes In vitro culture conditions for colony formation of human fresh peripheral T-cells using PHA and recombinant Interleukin-2 are defined. Peripheral lymphocytes, from six individuals, were exposed to X or gamma rays in vitro, and dose-survival curves were obtained. The results showed typical sigmoid curves similar to those observed when other mammalian cells are exposed to radiation. The D10 (dose required to kill 90% of the cells) was found to be 3.0 to 3.5 Gy.

Increased radiosensitivity of a subpopulation ot T-lymphocyte progenitors from patients with Fanconi's anemia.

In vitro radiation survival of peripheral blood T lymphocytes was studied in 15 clinically normal adults and 4 patients with Fanconi's anemia. Tritiated thymidine incorporation in a whole blood lymphocyte stimulation test (LST) and a newly developed whole blood T-lymphocyte colony assay were used to measure lymphocyte blastogenesis and colony formation in response to phytohemagglutinin (PHA) or...

Loss of interleukin-2 requirement for the generation of T colonies defines an early event of human T-lymphotropic virus type I infection.

Accessory cells and/or soluble factors, together with interleukin 2 (IL2), are required for the proliferation and differentiation of phytohemagglutinin (PHA)-activated T lymphocytes. Human T-lymphotropic virus, type I (HTLV-I), a human retrovirus isolated from patients with adult T cell leukemia, can transform T cells in vitro. We investigated the role of HTLV-I-transformed T cell lines as acce...

The regulatory role of interleukin 2-responsive T lymphocytes on early and mature erythroid progenitor proliferation.

To analyze the role of T lymphocytes in human erythropoiesis, we evaluated the effect of recombinant interleukin 2 (IL 2) on marrow CFU-E and BFU-E colony formation in vitro. IL 2 resulted in an increase in CFU-E and BFU-E colony numbers in a dose-dependent manner. This increase could be prevented by anti-Tac, a monoclonal antibody to the IL 2 receptor. Moreover, anti-Tac on its own resulted in...

Growth of clonogenic myeloblastic leukemic cells in the presence of human recombinant erythropoietin in addition to various human recombinant hematopoietic growth factors.

The effects of human recombinant erythropoietin (rEpo) in the presence of other stimulators on the growth of clonogenic leukemic blast cells from ten Japanese patients with acute myeloblastic leukemia were studied with an in vitro leukemic blast colony assay in methylcellulose culture. With the addition of rEpo alone, no leukemic blast colony formation was stimulated in any of the cases examine...

In Vivo Production of Interleukin-5, Granulocyte-Macrophage Colony-Stimulating Factor, Macrophage Colony-Stimulating Factor, and Interleukin-6 During Intravenous Administration of High-Dose Interleukin-2 in Cancer Patients

Recombinant human interleukin-2 (IL-2). administered t o cancer patients by continuous intravenous (IV) infusion (3 x 1V U/m’/d), was found t o induce the in vivo production of colony-stimulating factors (CSF). Plasma obtained from patients during IL-2 treatment stimulated in vitro colony formation of normal human bone marrow cells, depleted of mononuclear phagocytes and T lymphocytes. This col...
